Tasmanian Richie Porte was first to cross the Stage Five finish line at Willunga Hill for Team Sky
Porte completed the 151.5km course in three hours and 42 minutes.
Simon Gerrans will wear Ochre Jersey after his time bonus for finishing third has put him in the lead by one second.
The course started in McLaren Vale and featured two punishing climbs of Willunga Hill.
42-year-old Jens Voigt, the oldest rider in the Tour Down Under, won the first climb after being in the lead group of riders for most of the course.
The result has set up for an exciting final stage.
The riders will complete 20 laps of a 4.75km circuit around the streets of the Adelaide CBD, starting at 12.30pm.
The course starts and finishes on Bartels road, goes along Flinders street and will pass the Tour Village in Victoria Square.
